In this work, PhD student F. Giobbe from the University of Chalmers demonstrates that the skin of cockroaches is not only a major battleground between the roach’s parasites, but also contains arthropod immune defense cell “memory.”
Giobbe first showed a correlation between the outermost layer of the cockroach’s cuticle and the cells responsible for immune defense by using a fluorescent stain. He then compared this protein source in the roach to that of white blood cells in their environment. This was done by constructing a non-vaccine roach pupa. Mature cockroaches lack a fully developed immune system in their pupae, thus the white blood cells of newly emerged roaches cannot be used to assess the influence of immune defense in cockroaches’ cuticle. Still, he used a fluorescent stain to see if the bacterial and fungal cells are present on the outermost layer.
“Our results show the presence of cell, suggesting the deposition of cells into the cockroach skin hemolymph,” Giobbe said. Giobbe’s study provides evidence suggesting the cuticle might act as an immune response mechanism for immune defense.
The group have now begun to investigate and test the inner workings of the immune system by extracting the cells of the cuticle. Next, the researchers plan to compare the immune responses of cockroach hemolymph cells to that of the bacterial cells which are found on the cuticle. They will also test whether the hemolymph cells have any ability to fight against bacteria, something researchers have been looking for in the field.
A team of researchers at the UCL, led by Prof. Magda Tuschl and Amanda Seidenfaden from the Institute of Structural and Molecular Biology, revealed the role of methylation in regulating the transcription factor FOXO3 and in the human DNA damage response, revealing a previously unknown DNA damage response after acute oestrogen exposure.
